APPBP1/UBA3 is the heterodimeric E1 enzyme that conjugates Nedd8 to protein substrates. Nedd8 is conjugated onto the catalytic cysteine residue of UBA3 by formation of a thioester bond in an ATP-dependent reaction. The activated Nedd8 is then passed on to an E2 (Ubc12) to be finally attached to a substrate. Nedd8 is mainly conjugated on Cullins to increase the activity of the Cullin – RING E3 Ub ligases.
Biotium NucSpot 470, 1000X in DMSO
NucSpot 470 is a cell membrane-impermeant green fluorescent DNA stain. It is virtually non-fluorescent in the absence of DNA, but fluoresces bright green upon DNA binding. While other green nucleic acid stains like TOTO, TO-PRO, or SYTOX dyes stain both the nucleus and cytoplasm, NucSpot 470 specifically stains the nucleus of fixed and permeabilized cells. It also can be used to selectively stain dead cells in living cultures. NucSpot 470 has an excitation/emission of 460/546 nm and can be imaged using standard settings for FITC. Properties: Ex/Em= 460/546 nm (with DNA); Store at 4°C and protect from light.
Biotium DMAO, 2mM in DMSO
DMAO is a membrane-permeable DNA dye, excellent for staining live bacteria. Stained cells can be detected by fluorescence microscopy or flow cytometry. DMAO preferentially binds to dsDNA with green fluorescence (Ex/Em, DNA-bound = 496/528 nm). The dye is suitable for both gram-positive and gram-negative strains. Unlike the SYBR dyes, DMAO is extremely stable and can be routinely handled and stored at room temperature without degradation. Dihydroethidium (also called hydroethidium) is the chemically reduced form of the commonly used DNA dye ethidium bromide. The probe is useful to detect oxidative activities in viable cells, including respiratory burst in phagocytes. The LAMP fluorescent dye is an intercalating dye that can be used to track a LAMP reaction in real-time by fluorescence measurement. It is provided at 50X concentration and is sufficient for at least 500 LAMP reactions (0.5 ul dye per 25 ul LAMP reaction). The SYBR/FAM channel of common real-time fluorimeters can be used to read the dye.
Biotium One-Step Blue Protein Gel Stain, 1X
One-Step Blue is a ready-to-use protein gel staining solution. It produces fast (5-60 min) protein staining in a single step without fixation or washing. Proteins can be detected by visible blue staining, or by near-infrared fluorescence. In addition to rapid results and simple staining, One-Step Blue offers safer handling and disposal compared to Coomassie staining because it is entirely aqueous-based, without hazardous methanol or acetic acid. One-Step Blue has comparable sensitivity as the widely used Coomassie Blue, with a lower limit of detection around 10-20 ng depending on the detection method used. The staining is fully compatible with mass spectrometry and Edman-based sequencing.
